Spectrometry of Masses

MALDI-TOF

It MALDI-TOF work in the analysis of weighty molecules molecular one elevated for achieving the composition elementary. The technique MS/MS helps us in the structural characterisation of unknown compounds and in the analysis specific to biological samples.

Job request

In order to request the analysis of a sample for MALDI-TOF, you have to get yourselves with the staff responsible for the team in touch.

You will need to give the toxicity and any storage precautions. Once the report has been delivered, if you do not collect the sample within one week, any remaining sample will be destroyed.

How does it work?

The spectrometers of masses he STRIVES they generate ions in gas phase through the vaporisation induced by the laser of a solid mixture of sample and matrix in which this last acts as acceptor of energy of the laser that I transmit it in the sample, and produces the sublimation of the ions of the matrix and the sample. The ions are speeded in an electrical field for afterwards to penetrate in a tube of free flight. For a certain voltage of acceleration the time of flight (TOF) of the ion on arriving at the detector is proportional to its relation too much/load (m/z). The two parts of this instrument work in conditions of high emptiness. Increasing the power of the laser slightly a considerable fragmentation of the anàlit is produced and the ions evolve in the region of free itinerary, this process is called PSD. The màssica resolution incorporating a reflector in the end of the zone is improved of wants that it diverts the trajectory of the ions until finds a second detector with which structural information of the original molecule is achieved.

Applications / Practical cases

For its characteristics, the team is dedicated to the analysis of biomolecules (drugs, metabolites, oligosaccharides, oligonucleotides, peptides and proteins). With respect to the proteins, the MALDI-TOF can provide structural information at scale of too molecular, map of peptides and primary structure, sequences and posterior modifications.

  • Determination of the molecular weight of the intact protein.
  • Determination of the molecular ion in molecules of too small.
  • Obtaining of peptídic map (PMF) and identification of biological samples proteins (general practitioners, feeding, microbial crops, plants, etc) through MACOT, as an algorithm of search.
  • Identification of polypeptides.
  • Analyses of macromolecules, metabolites, drugs, polymers.
  • Identification of microorganisms for "Biotyper".
  • Direct analyses in plaques TLC.
