Area of research: Analysis of sperm quality

The objective analysis of the quality of ejaculated sperm, rich fraction or refrigerated or cryopreserved semen doses is a key element of guaranteeing the fertility and the prolificity of reproductive males.The main parameters that make up sperm quality are: sperm concentration, sperm morphology, sperm motility, sperm feasibility, the integrity of cell membranes, DNA fragmentation and sperm capacitation.To assess sperm concentration we have measuring devices equivalent to the spectrophotometers that evaluate objectively the number of spermatozoids per millilitre from the optic density of the sample.To determine sperm concentration, morphology and motility we use computer systems that allow us to objectively establish the number of spermatozoids per millilitre, the number and percentage of mature, immature and aberrant spermatozoids, and the various modalities of movement of spermatozoids (rapidly progressive, slowly progressive, circular, non progressive, immotile, etc.).To determine sperm viability, the integrity of the plasma, nuclear, acrosome and mitochondrial membranes, DNA fragmentation, sperm capacitation etc.we use specific fluorescent markers and we specifically assess all of these parameters, measuring the amount and type of fluorescence issued by flow cytometry.
