How does it work?

As in the case of the conventional DSC, in the microDSC the heat absorbed or given off is measured by a sample when this undergoes a programme of temperatures. However, the sensitivity of the microDSC is very superior to that of the conventional DSC, making her appropriate for the study of little energetic processes.

At instrumental/operational level, the main ones you differentiate between both techniques they are the way the flow of heat and the type of analysed sample are measured.

Unlike the conventional DSC of the UAT, the team of microDSC works according to the principle of “compensation of power”. The team has two independent ovens, one for the sample and another for the reference. The same programme of temperatures applies to both ovens, but when the sample experiences an exothermic or endothermic process, the moderate temperature will differ from the programmed one. The instrument will regulate the baked applied electrical power of the sample in order to maintain the same temperature in both ovens. This difference of respect power baked of reference will be directly the flow of moderate heat.

Biological samples are analysed by microDSC in dissolution. Unlike the case of the conventional DSC, where the samples are encapsulated, in microDSC the dissolution of sample as well as the reference fill directly in a capillary that acts as cell of measure.

Applications / Practical cases

The microcalorimetry is used widely in the study of biological samples, where the sensitivity of the classical DSC is not sufficient. Among others, it is they can quote the following applications:

• Studies of the union between ligands and proteins
• Characterisation of the stability of biomacromolecules
• Characterisation of the structure of proteins
• Study of lipids and interaction or changes of phase in lipídiques membranes
• Characterisation of nucleic acids, for example transitions B-Z in DNA
